Vascular Endothelial Redox Stress Mediated by Acrolein

Understanding the effect of a common environmental pollutant on the devleopment and prevention of cardiovascular disease in Wisconsin

Full Project Name:Vascular Endothelial Redox Stress Mediated by AcroleinPrincipal Investigator:Charles R. Myers, PhDAward Amount:$75,000
Award Date
July2009
Project Duration:24 months

Project Description Narrative:

Cardiovascular disease is a major cause of premature death and morbidity in Wisconsin. Atherosclerosis, the buildup of fats, cholesterol, and other substances on artery walls, is common in cardiovascular disease, and can have systemic effects that also affect the brain, kidneys, and circulation. The dysfunction of endothelial cells, cells that line the interior surface of blood vessels, can cause atherosclerosis.

Through this award, investigators aim to study how of acrolein, a common environmental pollutant produced by car exhaust and cigarette smoke, can damage endothelial cells.

Outcomes & Lessons Learned:

  • Examined the effects of acrolein on thioredoxin reductase (TrxR) which is essential for cell growth and survival
  • Discovered that acrolein irreversibly inhibits TrxR activity in endothelial cells. Widespread downstream effects that promote endothelial cell death are expected from this inhibition
  • Disseminated findings via a poster presentation and published two scientific papers
